I’m building a middleware platform for an existing system and was thinking of using Zapier.

The goal would be to charge those using my app based on usage.

Pricing TBD, but let's say it's $0.01 per call. Is this possible when building out an app on zapier?

Zapier is a 3rd party software program that helps you automate your daily task. So it’s up to you weather to charge for you service.
